  • Effect of Glucosamine Hydrochloride Capsules and Calcitonin on Matrix Metalloproteinase-3 and Osteopontin Level in Patients with Postmenopausal Knee Osteoarthritis

    目的 探讨盐酸氨基葡萄糖联合降钙素对绝经后膝骨关节炎基质金属蛋白酶3(MMP-3)和骨桥蛋白(OPN)表达的影响。 方法 2012年1月-6月将120例绝经后膝骨关节炎妇女随机分为盐酸氨基葡萄糖组(A组)、盐酸氨基葡萄糖+依降钙素组(B组)、依降钙素组(C组),每组40例,采用酶联免疫吸附试验测定各组血清MMP-3、OPN、雌二醇、Ⅰ型胶原C端肽(CTX)和Ⅰ型胶原N端前肽(PINP)水平。 结果 A组和B组在治疗后2周和6周其膝关节评分和视觉模拟评分明显优于C组(P<0.05),A组在治疗后2周MMP-3的表达改善明显(P<0.05),优于其他两组。治疗后6周,B组OPN表达水平改善明显(P<0.05),优于其他两组。C组和B组CTX和PINP水平明显改善(P<0.05),优于A组。 结论 盐酸氨基葡萄糖联合降钙素能有效改善绝经后膝骨关节炎的症状,可能通过调节MMP-3和OPN的复合体表达,实现改善关节软骨功能的目的。

  • Expression and Significance of E-cadherin, MMP-2 Protein, and MMP-9 Protein in Gastric Cancer Tissues

    Objective To explore the expressions of E-cadherin, matrix metalloproteinases-2 (MMP-2) protein,and matrix metalloproteinases-9 (MMP-9) protein in gastric cancer tissues, and to analyze the possible statistical relati-onship between the expressions of E-cadherin, MMP-2 protein, and MMP-9 protein, and clinicopathological features ofgastric cancer. Methods The ABC immunohistochemical staining was adopted to examine the expressions of E-cadherin,MMP-2 protein, and MMP-9 protein in 40 paraffin slices of gastric cancer (gastric cancer group), with the adjacent tissue as the control group (adjacent tissue group). The positive rates of 3 kinds of protein were compared between the2 groups, in addition, the statistical relationship between the expressions of the 3 kinds of protein and clinicopathological features of gastric cancer was examined respectively by SPSS 19.0 software. Results The expressions of E-cadherin, MMP-2 protein, and MMP-9 protein were all found in gastric cancer tissues and adjacent tissues. In gastric cancer tissue group, the expression of E-cadherin downregulated while the expressions of MMP-2 protein and MMP-9 protein upregulated in comparison to adjacent tissue group (P<0.05). The significant association was found between the expre-ssion of E-cadherin and the gastric cancer tissues of T3+T4 stage, N1-N3 stage, and Ⅲ+Ⅳ stage, which had lower positive expression rate (P<0.05). The expression of MMP-2 protein in gastric cancer tissues of M1 stage and Ⅲ+Ⅳstage upregulated (P<0.05), and the expression of MMP-9 protein upregulated in gastric cancer tissues of T3+T4 stage,Ⅲ+Ⅳ stage, or lowlydifferentiated+undifferentiated (P<0.05). No significant relationship was found in other clinical-pathological features and 3 kinds of protein except aforementioned significant relationship (P>0.05). Conclusions In the development progress of gastric cancer, the E-cadherin may get involved in the mechanism of tumor invasion and lymph node metastasis, MMP-2 protein may get involved in the mechanism of distant metastasis, and MMP-9 protein may get involved in the mechanism of differentiation and tumor invasion. The examination of those 3 kinds of markers may play an role in the judgment of tumor stage and estimation of prognosis in gastric cancer clinically.

  • Expressions and Clinical Significance of PTEN, Fas/FasL and MMP-2 in Gastric Cancer

    Objective To study the expressions of phosphatese and tensin homolog deletedin chromosom ten (PTEN), Fas/FasL system and matrix metalloproteinnases-2 (MMP-2) in human gastric cancer. Methods Seventy-five cases of gastric carcinoma were selected from paraffin wax embodied specimens with full clinicopathological data, and another 15 cases of normal gastric mucosa specimens were selected as the control group. SP immunohistochemistry was used to measure the expressions of PTEN, Fas/FasL and MMP-2 in them. The data was statistically analyzed by χ2 test and relative analysis. Results The expressions of PTEN, Fas/FasL and MMP-2 were correlated with the lymphatic metastasis, degree of infiltration, clinical TMN stage and pathological histological differentiated degree of gastric cancer (Plt;0.05). PTEN was positive correlated with Fas/FasL (r=0.401, Plt;0.001). MMP-2 was negative correlated with Fas/FasL (r=-0.720, Plt;0.001). MMP-2 was negative correlated with PTEN (r=-0.336, Plt;0.001). Conclusion There is guidance meaning in testing the expressions of PTEN, Fas/FasL and MMP-2 in gastric cancer to estimate the prevention, diagnoses, therapy and prognosis of gastric cancer.

  • Relationship Between Chlamydia Pneumoniae and Abdominal Aortic Aneurysm

    Objective To investigate the recent studies about the relationship between Chlamydia pneumoniae and abdominal aortic aneurysm. Methods The current literatures about the relationship between Chlamydia pneumoniae and abdominal aortic aneurysm were reviewed. Results Chlamydia pneumoniae is one of the most important factors for the formation of abdominal aortic aneurysm since Chlamydia pneumoniae can cause abdominal aortic aneurysm through the metabolism of matrix metalloproteinases, the apoptosis of smooth muscle cells in the vessels and the chronic infection of the wall of the aneurysm. Conclusion There maybe a distinguishingly close relationship between Chlamydia pneumoniae and abdominal aortic aneurysm, and Chlamydia pneumoiae may take an important role in the development and progress of the abdominal aortic aneurysm.

  • Effects of Nimesulide on Expression of Matrix Metalloproteinase- 2 in Human Colonic Cancer Cell Lines

    【Abstract】Objective To investigate the effects of selective cyclooxygenase-2 (COX-2) inhibitor nimesulide on the proliferation of colon adenocarcinoma cells in vitro and the expression of matrix metalloproteinase-2 (MMP-2). Methods The human colon cancer cell lines HT-29 and HCT-116 were employed in the study, grouped as nimesulide group, DMSO control group and blank control group. After treatment with nimesulide, the inhibitory effect of nimesulide on the proliferation of cancer cells was quantified by MTT assay, and the expression of MMP-2 in the cells was detected by quantitative zymography. Results Nimesulide inhibited the proliferation of HT-29 and HCT-116 cells in time and dosedependent manners. The inhibitory effect on HT-29 cells was ber than that on HCT-116 cells. Nimesulide downregulated the MMP-2 expression in HT-29 cells, whereas the expression in HCT-116 cells remained unchanged. Conclusion Nimesulide can obviously inhibit the growth of colon cancer HT-29 cells with positive COX-2 protein, suggesting that nimesulide may downregulate the expression of MMP-2 by inhibiting the activity of COX-2.

  • Construction of Recombinant Adenovirus Vectors Carrying Antisense Matrix Metalloproteinase-2

    【Abstract】Objective To construct a recombinant adenoviral vector carrying antisense matrix metalloproteinase2 (MMP2) for use in the gene therapy to inhibit the invasiveness and migratory capacity of hepatocellular carcinoma (HCC) cell line HepG2 in vitro and in vivo models. Methods Total RNA was extracted from HCC, and then a 500 bp fragment at the 5′ end of human MMP2 cDNA was synthesized by polymerase chain reaction (PCR) and was reversely inserted into the multiclone site (MCS) of the shuttle plasmid pAdTrack-CMV,with the resultant plasmid and the backbone plasmid pAdEasy-1,the homologous recombination took place in the E.coli BJ5183 and the recombinant adenoviral plasmid carrying the antisense MMP2 gene was constructed and generated. The adenoviruses(Ad-MMP2AS) were packaged and amplified in the HEK 293 cells.Then the viral titer was checked by GFP. Results The recombinant adenovirus vector carrying antisense MMP2 was constructed successfully, the b green fluorescence was observed in HEK 293 cells under a fluorescence microscopy. The viral titer was 1×108/ml. Conclusion The recombinant adenovirus Ad-MMP2AS constructed by us could introduce the antisense MMP2 into HepG2 effectively,which would provide experimental basis for reversing the overexpression of MMP2 in HCC and for inhibiting the invasiveness and migratory capacity of HepG2 in vitro and in vivo models.

  • Expression of Vascular Endothelial Growth Factor and Matrix Metalloproteinase-9 in Hepatocellular Carcinoma

    ObjectiveTo investigate the relationship between the expression of vascular endothelial growth factor (VEGF) and matrix metalloproteinase9 (MMP9) and the recurrence or metastasis of hepatocellular carcinoma (HCC).MethodsThe expression of VEGF and MMP9 in 50 HCC tissues and in 30 normal liver tissues were examined by immunochemistry.ResultsThe positive rates of VEGF and MMP9 in HCC tissue were 86% and 68% respectively, and 53.3% and 33.3% respectively in normal liver tissue. The positive rates of VEGF and normal liver tissue (Plt;0.05). The correlation between expressions of MMP9 and VEGF was very significant (r=0.98,Plt;0.01). The positive rates of VEGF and MMP9 in HCC with metastasis were higher than that of HCC without metastasis.ConclusionVEGF and MMP9 are expressed cooperatively. Both of them play important role in the invasion and metastasis of HCC.

  • The Effect of Cigarette Smoking on Expression of Matrix Metalloproteinase-9 in Airway Epithelium of Rats

    Objective To investigate the effects of smoking intensity, duration and cessation on mRNA and protein expressions of matrix metalloproteinase-9 ( MMP-9) in tracheal epitheliumof rats, and the relationship between smoking or smoking cessation and airway remodeling in chronic obstructive pulmonary disease ( COPD) . Methods Forty Wistar rats were randomly divided into 5 groups, ie. a normal control group, a long termheavy smoking group, a short termheavy smoking group, a long termlight smoking group,and a smoking cessation group which was exposed to room air for 10 weeks after long term heavy smoking.The expressions of MMP-9 mRNA and protein in tracheal epithelium of rats were detected by in situ hybridization and munohistochemistry respectively. Results ( 1) The pathological changes of emphysema were observed in the lung tissue of every smoking rat, and were most sever in the long term heavy smoking group. ( 2) Compared with the normal control group [ ( 0. 88 ±0. 88) PU, ( 2. 80 ±1. 66) PU] , the expressions of MMP-9 mRNA and proteins in tracheal epithelium were remarkable elevated in the long term heavy smoking group [ ( 22. 01 ±2. 86) PU, ( 20. 81 ±2. 46) PU] , the short term heavy smoking group [ ( 14. 94 ±3. 46) PU, ( 13. 68 ±2. 00) PU] , the long term light smoking group [ ( 6. 92 ±2. 71) PU,( 8. 84 ±1. 80) PU] and the smoking cessation group [ ( 19. 00 ±3. 36) PU, ( 14. 82 ±1. 74) PU] ( P lt;0. 01) . Compared with the long term heavy smoking group, the expressions of MMP-9 in tracheal epithelium were decreased in other three smoking groups ( P lt; 0. 05) . Conclusions Smoking could increase the expression of MMP-9 in tracheal epithelium and cause trachea damage and remodeling with intensity and duration in rats. Smoking cessation could decrease the MMP-9 expression and alleviate trachea remodeling,suggesting its role in the prevention of COPD.

  • Elevated Plasma sFas, sFas-L and MatrixMetalloproteinase-7 Levels in Sepsis and their Correlation with the Severity of Sepsis

    Objective To investigate the plasma levels of soluble Fas receptor ( sFas) , soluble Fas ligand ( sFas-L) and matrix metalloproteinase-7 ( MMP-7) and their correlation with disease severity as well as the prognosis of septic patients.Methods The plasma levels of sFas, sFas-L, sFas / sFas-L ratio and MMP-7 were measured by enzyme-linked immunosorbent assay and compared between32 patients with sepsis and 24 age and sex matched healthy controls. Based on the 28-day outcome, the patients were divided into a survival group and a death group. The difference in sFas, sFas-L, sFas/ sFas-L ratio and MMP-7 between the survival group and the death group were compared.Results Compared with the healthy control group, the concentration of plasma sFas, sFas-L and MMP-7 were significantly increased in the septic patients ( P lt; 0. 01) . Elevated plasma sFas and sFas-L were both positive correlated with the APACHEⅡ score and SOFA score. Although a modest negative correlation was found between plasma MMP-7 and APACHEⅡ score and SOFA score, but this correlation did not reach statistical significance ( P gt;0. 05) . The septic patients who died had significantly higher sFas-L level and lower sFas / sFas-L ratio as compared with those who survived ( P lt;0. 05) . Conclusion Plasma sFas, sFas-L and MMP-7 are associated with the disease severity and can serve as potential markers for predicting the outcome in septic patients.

  • The Effect of Smooth Muscle Cell Transplantation on MyocardialMMP2, MMP9 and TIMP3 Content One Week after Myocardial Infarction

    Objective To evaluate the effect of smooth muscle cell transplantation on myocardial interstitial reconstruction shortly after myocardial infarction. Methods A total of 48 female Wister rats were randomly divided into two groups with the random number table, the control group (n=24) and the smooth muscle cell transplantation group (n=24). The left coronary artery was ligated to set up the myocardial infarction animal model. An amount of 05 ml phosphate buffered saline(PBS) containing 1×106 smooth muscle cells or 0.5 ml PBS without cells was injected into the injured myocardium immediately. By immunoblot and reverse transcriptionolymerase china reaction (RT-PCR), we observed the amount of protein and mRNA of matrix metalloproteinase2(MMP-2), matrix metalloproteinase-9(MMP-9) and tissue inhibitor of metalloprotease-3 (TIMP-3) in the myocardium of the rats. Results The transplanted smooth muscle cells survived well. Compared with the control group, myocardial TIMP3 mRNA (1.06±0.22 vs. 0.81±0.19, t=-2.358, P=0.033) and protein content (3.33±0.53 vs. 1.63±0.47, t=-6.802, Plt;0.001) were significantly increased in the transplantation group. Myocardial MMP-2, MMP-9 mRNA (0.49±0.12 vs. 1.16±0.18, t=8.453, Plt;0.001; 0.45±0.12 vs. 0.80±0.11, t=5.884, Plt;0.001) and protein content (3.98±1.08 vs. 6.05±0.91, t=4.139, P=0.001; 0.39±0.14 vs. 0.57±0.17, t=2.409, P=0.031) [CM(1585mm]were significantly reduced in the transplantation group compared with the control group. Conclusion transplanted smooth muscle cells can survive well in the infarction myocardium and can increase the amount of myocardial TIMP-3 mRNA and protein content and reduce myocardial MMP-2, MMP-9 mRNA and protein content, which is an effective way to prevent harmful cardiac remodeling.
