  • Research Progress of Adjustment Mechanism between MicroRNA and Vascular Endothelial Cell Function

    Vascular endothelial cell(VEC) is a kind of simple squamous epithelium lined on the inner surface of blood vessels. VEC is an important barrier between the blood and tissue and it also plays a key role in regulating inflammation, thrombosis, endothelial cells mediated vasodilatation and endothelial regeneration. These processes should be controlled by a variety of complex mechanism which requires us to find out. With results of the researches in vascular endothelial cell function, the important roles that microRNA in vascular endothelial cell function draws more and more researchers' attention. MicroRNAs control gene expression in post-transcriptional level and affect the function of endothelial cells. This review focuses on the research progress on regulatory mechanism of microRNA to endothelial cell inflammation, thrombosis, vasodilation and endothelium regeneration.

  • The role of Interleukin-10 in Airway Inflammation in COPD Rats

    ObjectiveTo investigate the expressions of IL-10,tumor necrosis factor-α (TNF-α) and interferon-γ (IFN-γ) in serum and lung tissue of COPD rats in order to elucidate the potential mechanism of airway inflammation. MethodsForty-five healthy adult male SD rats were randomly divided into a COPD model group (n=30) and a normal control group (n=15). The COPD rat model was established by intratracheal instillation of lipopolysaccharide (LPS) and exposure to cigarette smoke for 28 days. The concentrations of IL-10,TNF-α and IFN-γ in serum and lung tissue were measured by ELISA. ResultsTNF-α level of serum and lung tissue in the COPD model group increased significantly compared with the control group(P<0.05),while the levels of IFN-γ and IL-10 decreased significantly[serum:(44.68±8.67) ng/L vs. (75.96±10.59) ng/L;lung tissue:(64.55±9.03) ng/L vs. (94.06±8.71) ng/L,P<0.01]. The level of IL-10 in serum and lung tissue was negatively correlated with TNF-α (serum:r=-0.67,lung tissue:r=-0.80,P<0.01). The level of IL-10 in serum and lung tissue was positively correlated with IFN-γ (serum:r=0.64,lung tissue:r=0.72,P<0.01). The level of IL-10 in serum and lung tissue was negatively correlated with the percentage of neutrophils(serum:r=-0.70,lung tissue:r=-0.67,P<0.01). ConclusionIn COPD rats,down regulation of IL-10 plays an important role in regulation of airway inflammation.

  • A Meta-analysis of the Relationship between Interleukin-1beta and Epileptic Seizure

    目的 系统评价白细胞介素1β(IL-1β)与癫痫发作的相关性。 方法 2012年5月-2013年10月检索PubMed数据库(1985年1月-2013年7月)、Medline数据库(1985年1月-2013年7月)、中国知网(1998年1月-2013年7月)和万方数据库(1996年1月-2013年7月),并辅以谷歌学术搜索引擎进行手工检索。由两名研究者按照研究的纳入与排除标准进行病例对照研究的选择,对入选文献的质量参照“非随机对照临床试验质量评价标准与计分表”进行评价和计分,评估纳入研究的方法学质量并对有效的信息进行数据提取。然后采用RevMan 5.0统计软件进行Meta分析,以加权均数差(WMD)为效应指标。 结果 共纳入5个研究,包括82例癫痫发作的患者和471例正常对照。所纳入的研究未描述详细的抽样方法及其检测方法的特异性。Meta分析结果显示:癫痫发作72 h以内的患者与正常对照人群之间血浆IL-1β的浓度差异无统计学意义[WMD=0.04 pg/mL,95%CI(−0.07,0.16)pg/mL,P=0.46]。 结论 IL-1β可能没有直接参与癫痫的发作,而是通过与其他炎性因子之间的相互作用来实现。因此,需要进一步的研究来证明IL-1β在癫痫发作中的作用。

  • Influence of N-Acetylcysteine on Burned Rats’ Inflammatory Effect

    目的:探讨N-乙酰半胱氨酸对烫伤大鼠炎症反应的影响。方法:48只Wistar大鼠随机分为实验组和对照组,制作烫伤休克模型,伤后1小时腹腔注射生理盐水40ml/kg抗休克,实验组于抗休克盐水中加入N-乙酰半胱氨酸160mg/kg,其后足量饮水。分别于烫伤前、烫伤后8小时、16小时及24小时,处死每组各6只大鼠并立即心脏取血,采用ELISA法检测大鼠血清中TNF-a含量,于创周近头侧 0.5cm处取皮肤全层组织,在显微镜下进行中性粒细胞计数。 结果:两组大鼠血清TNF-a含量伤后显著升高,8小时达到高峰,其后逐渐下降,两组各时点比较有显著差异(P<0.05);创周组织中性粒细胞计数于伤后随时间延长逐渐增多,两组比较有显著性差异(P<0.05)。 结论:N-乙酰半胱氨酸有助于降低烫伤大鼠血浆中TNF-a含量及创周组织中的中性粒细胞计数量,减轻烫伤大鼠的全身及局部炎症反应。

  • Inflammatoin,Malnutrition and Lipid metabolism in Maintenance Hemodialysis Patients

    目的:探讨尿毒症维持性血透(MHD)患者营养不良、炎症与脂质代谢紊乱三者间的相关关系,以及对心血管并发症的预测。方法:测51例尿毒症维持性血透患者血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、载脂蛋白(Apo)A1、ApoB、脂蛋白(a)LP(a)、白蛋白(ALB)、C反应蛋白(CRP)、血清铁蛋白(SF),与30例正常人进行对照研究。结果:MHD患者TG、LDL、ApoB、LP(a)、CRP、SF显著增高,HDL、ApoA1、ALB显著降低(P<0.05);随着透析时间的延长,各项检测指标变化进一步加重;且心血管事件发生者血清CRP和LP(a)明显增高,ALB明显降低,血脂紊乱更显著。相关性分析:ALB与HDL、ApoA1正相关,与CRP、LP(a)、TC、TG、ApoB负相关;CRP与LP(a)、TC、TG、ApoB、LDL、SF正相关,与HDL、ApoA1负相关,P均<0.05。结论:MHD患者存在明显的脂质代谢紊乱、炎症和营养不良,三者的相互促进和影响可能是心血管事件发生的危险因素。

  • Diagnosis and Treatment of Early Postoperative Inflammatory Small Bowel Obstruction after Laparoscopic and Open Resection for Colorectal Cancer

    目的 对比分析腹腔镜与开腹结直肠癌切除术后早期炎症性肠梗阻(EPISBO)的发生率,总结EPISBO的诊治经验。方法 非随机同期对照分析同一组医师连续实施的812例腹腔镜结直肠癌切除术(LS组)与615例开腹结直肠癌切除术(OS组)患者的资料,比较2组EPISBO发生率的差异。结果 术后EPISBO总发生率为4.56%(65/1 427),均经保守治疗治愈,无死亡病例。 LS组患者术后EPISBO的发生率为3.20%(26/812),低于OS组的6.34%(39/615),P<0.05。LS组患者术后EPISBO的治愈时间为(7.51±5.72) d,OS组为(8.12±5.74) d,2组差异无统计学意义(P>0.05)。结论 腹腔镜手术可降低EPISBO的发生率,EPISBO应以保守治疗为主。

  • Effect of ω-3 Polyunsaturated Fatty Acid on Inflammatory Response and Nutritional State of Patients with Gastrointestinal Malignancies after Operation

    Objective To research the effect of ω-3 polyunsaturated fatty acid (PUFA) on inflammatory response and nutritional condition after operation for patients with gastrointestinal malignancies. Methods Forty patients with gastrointestinal malignancies were included in this study from February 1st, 2009 to June 1st, 2009. Forty cases were randomly allocated to experimental group (20 cases) and control group (20 cases). Parenteral nutrition was conducted in continuous 7 days after operation. Comparing with control group, a dose of 10 g of ω-3 PUFA was given to experimental group every day in 7 days after operation in addition. Blood samples were gained before operation, the 2nd and 8th day after operation respectively to measure relative indexes about inflammatory response (WBC, neutrophilic granulocyte and C-reaction protein) and nutrition (total protein, albumin, prealbumin, siderophilin and lymphocyte). Reduction of body mass was also recorded. Results The baseline between experimental group and control group was comparable (Pgt;0.05). The levels of indexes about inflammatory response (WBC, neutrophilic granulocytem and C-reaction protein) and nutrition (total protein, albumin, prealbumin, siderophilin and lymphocyte) between experimental and control group did not reach statistically significant difference in the 2nd day after operation (Pgt;0.05). The levels of neutrophilic granulocyte and C-reaction protein in experimental group were lower than those of control group, and the level of lymphocyte in experimental group was higher than that of control group in the 8th day after operation, and all of them reached statistical significance (Plt;0.05). There was no statistical different in reduction of body mass between experimentalgroupandcontrolgroup.Conclusion ω-3 PUFA can depress the excessively inflammatory reaction and improve the nutritional condition of patients with gastrointestinal malignancies after operation.

  • Level and Significance of Inflammatory Cytokines in Patients with Acute Deep Venous Thrombosis of Lower Extremity

    Objective To study the significance of the levels of plasma inflammatory cytokines (IL-6,IL-8,IL-10 and TNF-α) in patients with acute deep venous thrombosis (DVT) of lower extremity. Methods Forty untreated DVT cases were selected as the subjects in the DVT group, while thirty healthy subjects, whose ages and genders showed no significant difference with the DVT patients, were collected as the control group. The plasma levels of IL-6, IL-8 and TNF-α were detected by radioimmunoassay (RIA), and the plasma level of IL-10 was measured by enzyme-linked immunosorbent assay (ELISA). Correlation analysis was used to investigate the relationships between the levels of different inflammatory cytokines within DVT group. Results The levels of plasma cytokines in the DVT group were all significantly higher than those in control group (P<0.001). The results of the correlation analysis showed that there were positive correlations between IL-6 and TNF-α (r=0.383, P<0.05), IL-10 and TNF-α (r=0.390, P<0.05), respectively, within the DVT group; whereas there were no correlations between IL-6 and IL-8, IL-6 and IL-10, IL-8 and IL-10, and IL-8 and TNF-α. Conclusion The levels of plasma cytokines increased significantly in patients of DVT. Inflammatory cytokines may play an important role in acute DVT by accelerating the pace of thrombosis, intensifying the inflammatory reaction around thrombus and aggravating the injured blood vessel.

  • Inhibition Effects of Recombination Human Growth Hormone and TPN on Stress Response of Postoperative Gastrointestinal Carcinoma Patients

    【Abstract】ObjectiveTo explore the mechanisms of anabolism intensified by recombination human growth hormone (GH) on the basis of total parenteral nutrition (TPN) during postoperative in gastrointestinal carcinoma patients. MethodsNinety-four gastrointestinal carcinoma patients undergone operation were randomly divided into TPN group and TPN+GH group. The levels of TNF-α, IL-1, IL-6 and CRP were detected in the first, third, seventh postoperative day. ResultsThe levels of TNF-α, IL-1, IL-6 and CRP were significantly lower in TPN+GH group than those in the TPN group at the first, third, seventh postoperative day (P<0.01). The levels of TNF-α, IL-1, IL-6 and CRP were significantly higher at the indicated time of postoperative days than the pre-operative days in the two groups (P<0.01). ConclusionBy inhibiting TNF-α, IL-1, IL-6 and CRP production in gastrointestinal carcinoma patients undergone operation and blocking high catabolism induced by inflammatory cytokines, GH promotes the synthesis of anabolism.

  • Inflammatory Pathogenesis in Pancreatic Cancer Development and Its Therapy Strategy

    【Abstract】Objective To investigate the relationship between the development of pancreatic cancer and inflammation, and the therapy strategy.Methods Related articles were reviewed.Results The pathogenesis of inflammation in pancreatic cancer development involves cytokines, NF-κB, COX-2, PPAR-γ, DNA damage, gene changes,etc. Based on these mechanisms some medications are under developing. Conclusion Accumulative effects of pancreatic inflammation may lead to DNA changes, and even pancreatic cancer development. Medications aimed at suppressing pancreatic inflammation may help with prevention and treatment of pancreatic cancer.
