  • The Effects of Atorvastatin Calcium on Vascular Endothelial Function of Patients with Carotid Artery Plague

    目的 探讨阿托伐他汀钙对颈动脉粥样硬化患者血管内皮的保护作用。 方法 选取2010年10月-2011年8月颈动脉粥样硬化患者80例,随机分为治疗组和对照组。对照组给予阿司匹林肠溶片0.1 g,早饭后口服1次;治疗组在此基础上给予阿托伐他汀钙20 mg,每晚口服1次,10个月为1个疗程。分别在治疗前后进行颈动脉彩色多普勒超声检测颈动脉中膜厚度及颈动脉粥样硬化斑块面积,测定甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、一氧化氮(NO)、血管内皮素-1(ET-1)水平。两组患者在1个疗程治疗结束后停药12周,测定TG、TCH、LDL-C、HDL-C、NO、ET-1水平。 结果 与治疗前比较,治疗组患者的TG、TC、LDL-C、ET-1水平显著降低,HDL-C、NO水平显著升高(P<0.01)。颈动脉中膜厚度和颈动脉粥样硬化斑块面积明显变小(P<0.05)。对照组无明显变化。治疗组停药12周后与停药时比较,TG、TC、LDL-C、ET-1水平显著升高,HDL-C、NO水平明显降低(P <0.05)。对照组无明显变化。 结论 阿托伐他汀钙能显著改善颈动脉粥样硬化斑块患者的血管内皮功能、血脂水平,稳定颈动脉粥样硬化斑块,促进斑块逆转,且需要长期坚持服用。

  • 中药与骨肉瘤耐药逆转研究探讨

    【摘要】肿瘤多药耐药(multidrug resistance,MDR) 是制约肿瘤成功化疗的重要因素之一,逆转MDR治疗恶性肿瘤是肿瘤治疗的有效策略,但由于肿瘤MDR机制复杂,耐药逆转药物作用靶点及机制单一,其治疗效果并不理想。中药抗肿瘤及耐药逆转具有高效低毒、多靶点、多阶段性等作用特点及其他优点,可针对肿瘤多药耐药的多种机制进行有效逆转。MDR亦是骨肉瘤化疗的主要障碍,骨肉瘤及其耐药有其特殊性,根据骨肉瘤耐药机理及中药耐药逆转机理,中药有潜力开发成为低毒高效的骨肉瘤耐药逆转剂,可能成为骨肉瘤治疗的理想药物。

  • Joint Detection of hTERT mRNA and GGT mRNA-H Expressions in Peripheral Blood of Small Hepatocellular Carcinoma Patients and Its Clinical Significance

    ObjectiveTo investigate the expressions and clinical significance of human telomerase reverse transcriptase (hTERT) mRNA and γglutamyl transpeptidase mRNA-H (GGT mRNA-H) in the peripheral blood of small hepatocellular carcinoma (HCC) patients. MethodsThe expressions of hTERT mRNA and GGT mRNA-H were detected in the peripheral blood of thirty patients with small HCC by RT-PCR, eighteen patients with benign liver diseases, and twelve normal volunteers. ResultsThe positive rate of hTERT mRNA and GGT mRNA-H expression in patients with small HCC were 80.0% (24/30) and 46.7%(14/30), respectively. In patients with hepatitic cirrhosis the positive rate of hTERT mRNA expression was 33.3% (6/18), while the expression of GGT mRNA was not detected. Both the expressions of hTERT mRNA and GGT mRNA-H were negative in all normal volunteers. The combination analysis of hTERT mRNA and GGT mRNA-H expression achieved positive rate of 86.7% in the diagnosis of small HCC, which was significantly higher than the positive rate of AFP (26.7%), Plt;0.05. ConclusionThe hTERT mRNA and GGT mRNA-H are significantly expressed in small HCC patients, the combination analysis of hTERT mRNA and GGT mRNA-H seems to be useful in the early diagnosis of small HCC.

  • Detection of Spreading Scope of Rectal Cancer to Mesorectum with RT-PCR and Its Clinical Significance

    【Abstract】ObjectiveTo detect the spreading scope of rectal cancer to mesorectum by RT-PCR using carcinoembryonic antigen (CEA) mRNA as a marker and to investigate the excision scope of mesorectum in resection of rectal cancer. MethodsForty specimens from 40 rectal cancer patients who underwent curative operation was employed to detect the metastatic deposits scattered in the mesorectum by RT-PCR using CEA as a marker. ResultsNine of 40 (22.5%) specimens contained metastatic deposits scattered in the mesorectum. The metastasis was just within the range of 4cm mesorectum under the verge of tumor. The tumor spreading to mesorectum is correlated with Dukes stages,the infiltrated depth of bowel wall, tumor differentiation and tumor type(P<0.05), and is not correlated with the size of tumor and the level of CEA(Pgt;0.05). ConclusionThe excision of mesorectum should be within the range of 5cm under the verge of tumor in surgical management of rectal cancer.

  • Reversal of Multidrug Resistance Gene mdr1 of Drug-Resistant Human Hepatocellular Carcinoma Cells with Antisense Oligodeoxynucleotide in Vivo

    Objective To investigate the reversal of the multidrug resistant gene mdr1 in vivo by antisense oligodeoxynucleotide (ASODN) on the basis of study in vitro. Methods The cultured drug-resistant human hepatocellular carcinoma cells were injected under the skin of axilla to establish the tumor model of nude mice. mdr1 ASODN accompanied by Lipofectamine were injected locally and ADM was injected intraperitoneally. Control 1 and control 2 were locally injected by Lipofectamine and normal saline separately, and ADM was also injected intraperitoneally. Results As time went on the tumor size increased and from the 5th day on alterations were marked, tumor size in different time phase showed marked difference to the prior time phase with significant difference (P<0.05). Tumor size in group ASODN was marked smaller than that of other 3 groups after the 5th day (P<0.05),while tumor size of group control 1,2 and group SODN in different phase showed no significant difference (Pgt;0.05). The results suggested that SODN and Lipofectamine showed no marked effect on tumor growth of nude mice and ASODN had marked inhibition effect on tumor growth. Conclusion mdr1 ASODN can also reverse multidrug resistance of drug-resistant human hepatocellular carcinoma cells in vivo. After the treatment the tumor’s growth in nude mice will slow down in a range of time.

  • Experimental Study on Changes of Expression of AFP mRNA in Human Hepatocellular Carcinoma Tissues after Oral Xeloda Therapy

    【Abstract】Objective To explore the changes of expression of AFP mRNA in human hepatocellular carcinoma (HCC) tissues after oral Xeloda therapy.Methods Total RNA was extracted from HCC tissue samples collect after operation and nested reverse transcription polymerase chain reaction (RT-nested PCR) assay was performed to determine the expression of AFP mRNA in this study.Results The final product of AFP mRNA amplified by RT-PCR was 174 bp and by RT-nested PCR was 101 bp. The AFP mRNA is positive in 12 of 21 patients (positive rate 57.14%) amplified by RT-nested PCR assay in Xeloda treatment group which is much lower than control group: 18 of 20 patients (positive rate 90.00%),P<0.05.The serum AFP value of Xeloda treatment group 〔(23.2±12.8) μg/L〕 is much lower than that of control group 〔(39.6±24.3) μg/L〕 four weeks after operation (P<0.05). However, There was no difference between two groups in serum AFP value before operation.Conclusion Xeloda can effectively suppress the expression of AFP mRNA in human HCC tissues and lower it’s product serum AFP value.The clinical application of Xeloda in HCC patients deserve further study.

  • Expression and Significance of the Multidrug Resistance-Associated Protein Gene in Primary Hepatocellular Carcinomas

    Objective To study the expression and significance of multidrug resistance-associated protein (MRP) gene in hepatocellular carcinoma (HCC). Methods Reverse transcription polymerase chain reaction (RT-PCR) assay was used to detect the expression of MRP mRNA in 25 fresh specimens of the primary HCC and its surrounding liver tissues. Immunohistochemistry LSAB technique was adopted to test MRP in 60 HCC specimens. The drug sensitivity was also tested by flow cytometry.Results The positive expression rates of MRP mRNA and MRP protein in primary HCC were 44.00%(11/25) and 45.00%(27/60) respectively. All the intensity of expression was low, but significant higer than its surrouding liver tissues (P<0.05). The intensity and expression rate of MRP protein in 5 recurrent HCC had a tendency to increase. There was a correlation between the expression of MRP mRNA and MRP protein in 25 patients using RT-PCR and immunohistochemistry assay (Plt;0.05). Detected by flow cytometry, the average sensitivity of drugs in vitro of 60 HCC sp-cimens were 5-FU (15.80±7.63)%,DDP(18.45±9.59)%,ADM(17.95±7.99)%,MMC(16.60±8.69)% and CTX(17.40±10.14)%. Only 5FU and ADM were significantly affected by the expression of MRP protein (Plt;0.05).Conclusion The expression of MRP in primary HCC may be one of the important mechanisms of the intrinsic and acquired drug resistance in HCC. To study the expression of MRP could give a predictive value in HCC chemotherapy.

  • REVERSAL OF MULTIDRUG RESISTANCE OF HUMAN HEPATOMA RESISTANT CELLS IN VITRO BY ANTISENSE PHOSPHOROTHIOATE OLIGONUCLEOTIDE

    Objective To investigate the reversal effect of antisense phosphorothioate oligonucleotide (ASOND) on human hepatoma resistant cells. Methods Human hepatoma resistant cells SMMC-7721 was transfected with synthetic antisense phosphorothioate oligonucleotide complementary to the 5′ region flanking the AUG initiation codon mediated by lipofectamine. In vitro drug sensitivity was measured by MTT assay. The expression of P-170 was determined by flow cytometry and mRNA was assessed by RT-PCR. Results ASOND inhibited the expression of mRNA and p-170 in SMMC-7721, enhanced the sensitivity of SMMC-7721 to chemotherapeutic drug. The best inhibitory effect was achived by the dose of 0.5μmol/L. Conclusion ASOND enhanced the sensitivity of SMMC-7721 to chemotherapeutic drug and reversed the multidrug resistance of SMMC-7721 partially.

  • THE ALTERNATION OF AFP-mRNA DETECTED IN BLOOD DURING LIVER RESECTION FOR HEPATOCELLULAR CARCINOMA AND ITS SIGNIFICANCE

    【Abstract】Objective To investigate whether liver resection for hepatocellular carcinoma (HCC) causes dissemination of liver tumor cells into blood circulation. Methods Fourteen patients with HCC, but without evidences of metastasis, were enrolled for the study. Blood samples of peripheral blood before skin incision and after abdominal wall suture, and of hepatic venous blood and portal venous blood after liver parenchyma dissection, were obtained. AFPmRNA was detected by reverse transcription polymerase chain reaction assays, the change of the level of its expression during operation was assessed by semi-quantitative analysis. Results The rate of its expression before and after operation in peripheral blood, and during operation in portal venous blood and in hepatic venous was 42.9%, 35.7%, 42.9% and 57.1% respectively. There were no differences between them. However, the level of its expression in hepatic venous blood was significantly higher than others (P<0.05). Conclusion Liver resection for HCC induces releases of cells from the liver, probably including tumor cells, into blood circulation.

  • OVEREXPRESSION OF THE MDRl GENE EXPRESSION IN HUMAN PRIMARY HEPATOCELLU-LAR CARCINOMAS

    We have devised a highly sensitive, specific, and quantitative assay for multidrug resistance (mdr1) mRNA expression based on the reverse transcription-polymerase chain reaction (RT-PCR). mdr1 mRNA levels were detected in 30 human primary hepatocellular carcinoma (PHC) tissue and adjacent liver tissue. Five of the patients had received chemotherapy before hepatectomy. The results show that the level of expression of mdr1 gene is higher in tumor tissue than in adjacent liver tissue. mdr1 gene is overexpressed in PHC after chemotherapy. Furthermore, mdr1 gene expression in the treated tumor adjacent liver tissue is higher than that in untreated tumor adjacent liver tissue. Our results indicated that overexpression of mdr1 gene may be responsible for the intrinsic and acquired drug resistance of PHC.
