  • Effect of Radix Salviae Miltiorrhizae on Patency and Proliferation Lesion of Autologous Vein to Artery Grafts in the EarlyMiddle Stage

    ObjectiveTo assess the effects of Radix Salviae Miltiorrhizae (RSM) on patency and proliferation lesion of autologous vein to artery grafts in the earlymiddle stage.MethodsAutologous jugular vein was grafted into abdominal artery in the rats. The rats were divided into two groups: RSM group and control group. The rats in RSM group were fed with RSM [24 g/(kg·d )],which began 1 day before operation and continued until harvesting. Vein grafts were harvested at 1,3 days, 1, 2, 4 and 8 weeks after surgery for examining the patency, thickness of intimamedia and expression of proliferating cell nuclear antigen (PCNA). ResultsNo significant differences existed in patency of vein grafts between the two groups (Pgt;0.05). The intimamedia thickness of the vein grafts in RSM group decreased 1/3 compared with control group at 2, 4 and 8 weeks (P<0.01). The PCNA positive cells in RSM group reduced significantly as compared to the control group (P<0.01). ConclusionRSM can inhibit proliferation lesion of vein grafts but has no influence on patency of vein grafts in the earlymiddle stage.

  • Effect of Endothelin and Nitric Oxide Changes in Local Site of Vein Transfer with Delayed Breaking Pedicle in Rabbits

    Objective To study the changes of endothelin (ET) and nitric oxide (NO) in the local site of vein transfer with delayed breaking pedicle and the relation with vasospasm and vein transfer in rabbits. MethodsThe ET concentration of blood was determined with the radioimmunoassay method. The plasma NO-2,NO-3 levels in the local site of vein transfer with delayed breaking pedicle, which reflected NO levels indirectly, were detected with Ultravioletvisible (UvVIS ) spectrophotometer. ResultsThe endothelin concentration of blood was increased significantly at 2, 4 hour after the operation (P<0.01), and at 8 hour after the operation (P<0.05). The plasma NO level was significantly decreased at 2, 4 hour after the operation (P<0.01). But at 24 hour after the operation, the plasma NO level was increased significantly (P<0.05). Conclusion The recovery of ET concentration of blood and the increase of plasma NO at 24 hour after the operation are the cause of the reduced incidence of vascular crisis of vein transfer with delayed breaking pedicle, and the very time point is the optimum moment for pedicle breaking.

  • The Role of Low Power Red Laser Illumination on Neointimal Formation in Vein Graft Model

    ObjectiveTo evaluate the effect of low power red laser illumination on the intimal proliferative response in vein graft models.MethodsAutogenous vein graft models were established in 80 rats by transplanting jugular vein to carotid artery by end to end anastomosis, and were randomized into two groups: control group (graft nonilluminated), laser illumination group (0.9 J/cm2).The grafted veins were harvested at 3,7,14 or 28 day respectively after operation. IH (intimal hyperplasia) and SMC (smooth muscle cell) proliferations were pathologically and immunohistochemically observed and analyzed by computer digitizing system. ResultsThere were no significant differences in the intimal average thickness and the areas of lumen between two groups for 3 day. Laser group was significantly lower than the control in both the intimal average thickness and the stenosis of lumen at 7 day,14 day and 28 day (P<0.05).Immunohistochemical analysis of PCNA indicate the decreased positive cell in laser group compared with the control group (P<0.01).ConclusionThese preliminary results demonstrate that a certain density of low power red laser illumination in vein graft inhibits SMC proliferation and neointimal hyperplasia in rat.

  • 大鼠静脉移植模型建立的常见并发症防治

    目的 探讨大鼠自体静脉移植模型建立常见并发症的预防和处理措施。 方法 采用两点定位法,将65 只大鼠的右侧颈外静脉分支与同侧颈总动脉断端行间断全层外翻端端吻合。通过吻合后血管充盈和搏动情况及远端勒血实验来确定吻合口通畅。从术中操作和术后护理的角度,分析常见并发症的发生原因及相应的防治措施。 结果 65 只大鼠自体颈静脉移植,52 只实验成功,成功率80.0(52/65),其中术中死亡率7.7%(5/65),术后死亡率12.3%(8/65)。导致大鼠术中死亡的主要原因有麻醉意外、呼吸道分泌物阻塞窒息、触碰迷走神经突发心搏骤停、失血性休克;术后死亡的主要原因有吻合口漏血、血管空气栓塞、吻合口血栓形成或闭塞、感染。 结论 实验过程中熟悉各种并发症发生的原因和防治措施,提高手术技巧,可减少并发症的发生,提高手术成功率,达到预期的模型建立效果。

  • Experimental Study of External Stents Preventing Porcine Saphenous Vein Grafts Restenosis

    Abstract:Objective To evaluate the effect of external stents on preventing vein graft neointima formation and medial thickening with non-restrictive macro porous polyester stent around porcine vein grafts. Methods Studies were performed by using "white race" pigs (n= 10) weight 25-30 kg. All the animals underwent bilateral saphenous vein into carotid artery bypass grafting. In each animal, a maeroporous stent was placed around a graft on one side and a control (unstented) graft on the opposite side. The polyester stent was shaped to cover both anastomoses completely. The size of the stem allowed unrestricted expansion of the graft in initial response to arterial pressure. After 35 days of surgery,all animals were taken to remove the grafts. Graft wall dimensions, platelet- derived growth factor (PDGF) expression and cell proliferation using proliferating cell nuclear antigen (PCNA) were measured on histological sections. Results Stents significantly reduced neointimal thickening (0. 4872 ± 0. 0706 mm vs. 0. 2259± 0. 0553mm,P〈0. 01)and medial thickening (0. 6246±0. 0859mm vs. 0. 4201±0. 0615mm,P〈0. 01). Stents significantly reduced the percentage of cells expressing PDGF and PCNA. Media, intimal PCNA index was reduced from 7. 980/00± 4. 060/00 to 3.35±0.95%(P〈0.01), PDGF index was reduced from 9.47%±5.35% to 2.67%± 0.97% (P 〈0. 01). Conclusion External non-restrictive polyester stent can significantly inhibit neointimal formation and medial thickening, and may prevent late vein grafts restenosis.

  • Human Heme Oxygenase-1 Gene Transfection Inhibits Intimal Hyperplasia of Vein Grafts in Rabbits

    ObjectiveTo evaluate the effect of human heme oxygenase 1 augmentation in vein grafts by adenoviral mediated gene transfer of heme oxygenase 1 (Ad hHO 1) on intimal hyperplasia.MethodsTwenty one Japanese white rabbits were divided into three groups: control group, Ad null control group, and Ad hHO 1 group(each group 7 rabbits). During the operation of rabbits jugular vein into carotid artery interposition grafting, harvested rabbit jugular vein segments were exposed for 30min at room temperature to heparin saline, recombinant replication deficient adenovirus encoding hHO 1(Ad hHO 1, 1× 10 9pfu/ml), and nude recombinant replication deficient adenovirus (Ad null, 1×10 9pfu/ml). Quantitative histological studies of the vein segments were performed 28 days after operation. Protein of hHO 1 was detected with method of immunohistochemical staining(S P) in 14 days and 28 days after operation.ResultsThe average intimal thickness, medial thickness and intimal to medial(I/M) ratio were calculated for each group 28 days after bypass operation. Compared to intimal thickness, I/M ratio of control group veins and Ad null group veins,Ad hHO 1 group veins decreased significantly( P lt;0.01). There was no statistically difference in medial thickness ( P gt;0 05). Strong staining of hHO 1 was detected in vein grafts wall of Ad hHO 1 group.ConclusionAd hHO 1 gene therapy may inhibit intimal hyperplasia of vein grafts in rabbits.

  • eNOS基因转染预防静脉移植血管再狭窄

    摘 要: 目的 应用含牛内皮型一氧化氮合成酶(eNOS)基因重组腺病毒(Ad5CMVNOSⅢ)转染静脉移植血管、观察eNOS基因预防静脉移植血管再狭窄的作用。方法 将21只杂种犬分为3组,手术对照组、Ad5CMVLac—Z(含大肠杆菌β半乳糖苷酶基因重组腺病毒)对照组和Ad5CMVNOSⅢ干预组。在犬颈静脉、颈动脉旁路血管移植术中分别应用Ad5CMVNOSⅢ病毒液或Ad5CMVLac—Z病毒液常温浸泡法感染静脉移植血管30分钟,术后28天病理切片观测移植血管新内膜增生状况。结果 与正常犬颈外静脉相比,手术对照组、Ad5CMVLac—Z对照组和Ad5CMVNOSⅢ干预组颈外静脉移植血管内膜/中膜比较均有不同程度增加(P<0.05),但Ad5CMVNOSⅢ组内膜/中膜比显著低于另外2个对照组(P<0.05),新内膜增生明显减轻。结论 Ad5CMVNOSⅢ感染静脉旁路移植血管对预防再狭窄有一定作用。

  • 大鼠自体静脉移植后内膜增生中Ⅰ、Ⅲ型前胶原mRNA及其蛋白的早期表达

    目的 探讨静脉移植后内膜增生早期的发生机制.方法 采用免疫组织化学、核酸原位杂交等方法,对大鼠静脉移植后7天、14天、28天和42天内膜增生中的Ⅰ型、Ⅲ型[α1(Ⅰ)、α1(Ⅲ)]前胶原Mrna及Ⅰ型、Ⅲ型胶原蛋白的表达进行定位、半定量观察.结果 与组织修复相关的胶原合成于术后7天出现,术后14天消失.术后14天起,α1(Ⅰ)、α1(Ⅲ)前胶原Mrna开始大量在增生内膜的平滑肌细胞中表达,并于术后28天达高峰(Plt;0.05),与相应胶原的表达呈同步增长.结论 静脉移植后7天Ⅰ型、Ⅲ型胶原即开始在内膜增生中的平滑肌细胞内转录及合成,且与平滑肌细胞的增殖无关.提示防治静脉移植后内膜增生至少应从术后14天开始,并应包括抑制细胞增殖及胶原合成两个方面.

  • PRELIMINARY STUDY ON EFFECTS OF HUMAN BRAIN-DERIVED NEUROTROPHIC FACTOR GENE-MODIFIED BONE MARROW MESENCHYMAL STEM CELLS BY INTRAVENOUS TRANSPLANTATION ON STRUCTURE AND FUNCTION OF RAT INJURED SPINAL CORD

    Objective To transplant intravenously human brain-derived neurotrophic factor (hBDNF) genemodified bone marrow mesenchymal stem cells (BMSCs) marked with enhanced green fluorescent protein (EGFP) to injured spinal cord of adult rats, then to observe the viabil ity of the cells and the expressions of the gene in spinal cord, as well as theinfluence of neurological morphological repairing and functional reconstruction. Methods Ninety-six male SD rats weighing (250 ± 20) g were randomly divided into 4 groups: hBDNF-EGFP-BMSCs transplantation group (group A, n=24), Ad5-EGFPBMSCs transplantation group (group B, n=24), control group (group C, n=24), and sham operation group (group D, n=24). In groups A, B, and C, the spinal cord injury models were prepared according to the modified Allen method at the level of T10 segment, and after 3 days, 1 mL hBDNF-EGFP-BMSCs suspension, 1 mL Ad5-EGFP-BMSCs suspension and 1 mL 0.1 mol/L phosphate buffered sal ine (PBS) were injected into tail vein, respectively; in group D, the spinal cord was exposed without injury and injection. At 24 hours after injury and 1, 3, 5 weeks after intravenous transplantation, the structure and neurological function of rats were evaluated by the Basso-Beattie-Bresnahan (BBB) score, cortical somatosensory evoked potential (CSEP) and transmission electron microscope. The viabil ity and distribution of BMSCs in the spinal cord were observed by fluorescent inverted phase contrast microscope and the level of hBDNF protein expression in the spinal cord was observed and analyzed with Western blot. Meanwhile, the expressions of neurofilament 200 (NF-200) and synaptophysin I was analyzed with immunohi stochemistry. Results After intravenous transplantation, the neurological function was significantly improved in group A. The BBB scores and CSEP in group A were significantly higher than those in groups B and C (P lt; 0.05) at 3 and 5 weeks. The green fluorescence expressions were observed at the site of injured spinal cord in groups A and B at 1, 3, and 5 weeks. The hBDNF proteinexpression was detected after 1, 3, and 5 weeks of intravenous transplantation in group A, while it could not be detected in groups B, C, and D by Western blot. The expressions of NF-200 and synaptophysin I were ber and ber with transplanting time in groups A, B, and C. The expressions of NF-200 and synaptophysin I were best at 5 weeks, and the expressions in group A were ber than those in groups B and C (P lt; 0.05). And the expressions of NF-200 in groups A, B, and C were significantly ber than those in group D (P lt; 0.05), whereas the expressions of synaptophysin I in groups A, B, and C were significantly weaker than those in group D (P lt; 0.05). Ultramicrostructure of spinal cords in group A was almost normal. Conclusion Transplanted hBDNF-EGFP-BMSCs can survive and assemble at the injured area of spinal cord, and express hBDNF. Intravenous implantation of hBDNF-EGFP-BMSCs could promote the restoration of injured spinal cord and improve neurological functions.
