  • EXPERIMENTAL STUDY ON PROTECTIVE EFFECTS OF ANISODAMINE ON ISCHEMIA-REPERFUSION INJURY IN RAT LIVER

    【Abstract】Objective To study the protective effects of anisodamine on liver ischemia-reperfusion injury in rats. Methods One hundred and sixty male Wistar rats were randomly divided into the normal control (n=10), ischemiareperfusion (n=50), normal saline (n=50) and anisodamine (n=50, 2.0 mg/kg) groups. The animals were killed 1, 3, 6, 12, 24 hours after ischemia induced for 60 minutes and followed by reperfusion. Plasma endothelin-1 (ET-1), hyaluronic acid (HA), glutamic-pyruvic transaminase enzyme (ALT) were measured, and the hepatic histopathologic alterations were also observed. Results The plasma ET-1, HA and ALT concentrations were markedly increased after liver ischemiareperfusion.The hepatic congestion was significantly obvious. An intravenous injection of anisodamine before ischemiareperfusion could decrease the plasma HA and ALT concentrations and relieve the hepatic congestion. Conclusion Anisodamine can improve hepatic microcirculatory disturbances after reperfusion and have hepatoprotictive effects on rat liver ischemia-reperfusion injury.

  • The protection of aminoguanidine,silymarin and anisodamine on growth and cytosolic free calcium changes of retinal capillary pericytes cultured in glycation products

    Objective To investigate the protective effect of aminoguanidine(AG),silymarin (Sil) and anisodamine (Ani) on retinal capillary pericytes cultured in glycosylation products. Methods MTT cololrimetric assay, [3H] thymidine incorporating and fluorescent indicat or fura-2 acetoxy-methyl ester (Fura-2AM) were used to study the influence of AG,Sil and Ani on the growth,DNA synthesis,and cytosolic free calcium ([Ca2 ]i)changes of pericytes cultured in the medium contained early glycation products (EGs) or advanced glycation end products (AGEs). Results Cultured in the medium contained EGs,the A value by MTT assayed and amount of [3H] thymidine incorporating in AG group and Sil group were obviously elevated than those of control group(Plt;0.01);but the [Ca2 ]iconcentration in both groups were decreased significantly comparing with control group(Plt;0.01 and 0.05).Under the condition of AEGs,only AG group was distinctly increased on the A value and amount of [3H] thymidine incorporatin g (Plt;0.01),and [Ca2 ]i concentration was markedly decreased (Plt;0.05) comparing with control group. Conclusion AG has the portective effect on pericytes against the proliferative inhibition and excessive elevation of [Ca2 ]i concentration in cytosol which are induced both by EGs or AGEs.Silymarin has the effect for those only by Egs-induced.Ani has no protective effect no pericytes nei ther cultured in medium with EGs nor with AGEs. (Chin J Ocul Fundus Dis, 2001,17:192-194)

  • Experimental observation of cluster therapeutic regimen in early stage of blast-induced acute lung injury in rats

    ObjectiveTo observe the effects of cluster therapy combined with anisodamine, dexamethasone and ambroxol on arterial blood gas, inflammatory cytokines and pulmonary pathological changes by making an early (<48 h) primary blast lung injury model in rats. MethodsEighty Wistar rats were randomly divided into six groups, ie. a control group (n=5), an injury group (n=15), an ambroxol treatment group (n=15), a dexamethasone treatment group, a scopolamine treatment group (n=15), a combination of ambroxol, dexamethasone and anisodamine group (n=15). The treatment groups were injected intraperitoneally with ambroxol 46.7 mg/kg (three times a day) or (and) dexamethasone at 5 mg·kg–1·d–1 or (and) anisodamine at a dose of 3.33 mg/kg (three times a day). The rats in the injury group were injected intraperitoneally with an equal volume of normal saline. Respiratory rate and weight change were observed before and after injury. Five rats were sacrificed at 6 hours, 24 hours and 48 hours after injury in each experimental group. Arterial blood gas analysis, Yelverton pathological score, lung tissue wet/dry weight ratio, serum tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were measured. The lung histopathology was observed. ResultsAfter lung blast injury, the rats in the injury group showed progressive respiratory acidosis, and hypoxemia increased with the increase of IL-6 and TNF-α in a time-dependent manner. The PaO2 decreased in the groups with ambroxol, dexamethasone and anisodamine alone or in combination with anisodamine, and the contents of serum IL-6 and TNF-α decreased. Pathological edema and inflammatory infiltration of lung tissue were alleviated significantly. ConclusionsAfter treatment with dexamethasone, anisodamine and ambroxol after lung blast injury, blood gas analysis is improved, inflammatory factor level is decreased and lung injury is alleviated, indicating that the three drugs can treat lung detonation injury in rats. The cluster therapy is superior to the single drug therapy.
