  • Calcium-Binding Tyrosine Phosphorylation-Regulated Gene Characteristics and Significance in The Pathogenesis of Tumor

    Objective To summarize the traits of calcium-binding tyrosine phosphorylation regulate gene and the significance in tumor incidence. Methods The domestic and foreign literatures about calcium-binding tyrosine phosphorylation regulate gene involved in the regulation of signaling pathways and research status in a variety of tumors were reviewed. Results Calcium-binding tyrosine phosphorylation regulate gene induced the abnormal proliferation of cells through multiple mechanisms. There was closely relation between the occurrence of many tumors and abnormal expression of calcium-binding tyrosine phosphorylation regulate gene. In the distribution of different epithelial tumors, the pathway of calcium-binding tyrosine phosphorylation regulate gene involved in the regulation was same, and the effect target was similar. Conclusion Further study of the calcium-binding tyrosine phosphorylation regulate gene is expected to provide a new way for clarify the occurrence and development mechanisms of tumors, and can serve as important means of early diagnosis and adjuvant therapy.

  • Effect of 103Pd Radioactive Stent on Human Cholangiocarcinoma Cell Apoptosis and Expression of Fas Gene

    Objective To research the effect of γ-ray released from 103Pd radioactive stent on the expression of Fas gene and its relation with apoptosis of cholangiocarcinoma cell and the significances through the establishment of human cholangiocarcinoma model. Methods The model of nude mouse with implanted human cholangiocarcinoma was established. The mice were divided into study group and control group, 37 MBq 103Pd biliary stent was implanted in the study group and the ordinary metal biliary stent was implanted in the control group. The volume of tumor was measured, the cell apoptosis was detected by the TUNEL method and the expression of Fas gene of the cell apoptosis of the induced human cholangiocarcinoma was checked out by immunohistochemistry staining 10 d after the implantation. Results Compared with the control group, the growing speed of the volume of tumor in study group was significantly reduced (Plt;0.05), the expression positive rate of Fas gene was significantly higher (Plt;0.05), and the apoptotic rate of cancer cells was also higher (Plt;0.01). Conclusions The 103Pd radioactive stent can induce the cell apoptosis in nude mouse model with implanted human cholangiocarcinoma inhibit the cell growth of bile duct cancer and may promote the apoptosis of cancer cells by increasing the expression of Fas gene. It may be helpful for the further study of treatment for bile duct cancer using 103Pd radioactive stent.

  • Relation of Apoptosis Muscular Cell in 103Pd Radioactive Stent of Dog Biliary Muscular Formation and Inhibition of Biliary Ductal Stricture

    【Abstract】Objective To research relation of apoptosis muscular cell in 103Pd radioactive stent of dog biliary muscular formation and inhibition of biliary ductal stricture. Methods Twelve dogs were randomly divided into two groups, which were general stent group and 103Pd radioactive stent group. General stent and 103Pd radioactive stent were respectively put into extrahepatic biliary tract of two groups. After 30 days all dogs were killed, and biliary tract were taken out. Apoptotic cells were detected by immunohistochemical methodsand agar electrophoresis, and nucleus browyellow was positive cell. Dog biliary duct cross-sections were stained by hematoxylin-erosin; area and perimeter of lumen,thickness of inner membrane and stenosis degree in bile duct were analysed by image analysis software of computer.Results The apoptotic biliary duct smooth muscle cell [(87.9±7.96)/cm2] was more significantly increased in the 103Pd radioactive stent group than in the general stent group [(5.6±0.51)/cm2], P<0.05; and comparing with the general stent group, the 103Pd radioactive stent significantly reduced biliary muscular formation thickness. Conclusion The result shows that 103Pd radioactive stent can inhibit proliferation of biliary ductal smooth muscle cell.

  • Analysis of Risk Factors of Functional Delayed Gastric Emptying after Pylorus-Preserving Pancreatoduodenectomy

    Objective To investigate the risk factors and preventions of functional delayed gastric emptying (FDGE) after pylorus-preserving pancreatoduodenectomy (PPPD). Methods The clinical data of 41 patients after undergoing PPPD between 2003 and 2009 in this hospital were analyzed retrospectively.Results In all 41 cases, postoperative complications developed in 13 patients (31.7%), in which 7 patients developed FDGE (17.1%). The complications excluding FDGE (P=0.010) and diabetes (P=0.024) had remarkable relations with the FDGE in the univariate analysis; Compared with the non-FDGE patients, the albumin was declined obviously (P=0.020) while the serum direct bilirubin increased significantly (P=0.036) in the FDGE patients, while the development of FDGE had relation only with the albumin (P=0.039) and the complication of diabete (P=0.047) by the binary logistic regression analysis. Conclusion In the patients undergoing PPPD, preoperative control of the blood glucose, preoperative correction of hypoproteinemia and hyperbilirubinemia, and centralizing PPPD in high-volume have possibly positive significance for the prevention of FDGE.

  • Effect of 103Pd Radioactive Stent on The Expression of Fas Gene and Its Relation with Apoptosis of Bile Duct Cancer Cells

    Objective To research the effect of γ-radiation released from 103Pd radioactive stent on the expression of Fas gene and its relation with apoptosis of bile duct cancer cells lines. Methods  The cancer cells of bile duct were dissociated into suspension in culture flasks, and the number of cells was counted by hemacytometry. The suspension was then stored in 2 ml freezing tubes in the density of 1×105/ml. They were set in two stents: general stent (general stent group) and 103Pd radioactive stent (103Pd stent group). The expression of Fas gene and apoptosis of bile duct cancer cells in general stent group and 103Pd stent group were analysed with immunohistochemistry technique and TUNEL method, respectively. Results  The expression level of Fas gene in 103Pd stent group was significantly higher than that in general stent group (P<0.05), and the number of apoptotic cancer cells in 103Pd stent group was also significantly more than that in general stent group (P<0.01). Conclusion There is a correlation between the expression of Fas gene and the apoptosis of bile duct cancer cells, which means that 103Pd radioactive stent may increase the expression of Fas gene and promote the apoptosis of cancer cells. It may be helpful for the further study of treatment for bile duct cancer using 103Pd radiative stent.
