  • Expression of hepatocyte growth factor receptor in human epiretinal membranes and RPE cells

    Objective To investigate the expression of hepatocyte growth factor receptor (HGFR) in epiretinal membranes (ERM) of eyes with proliferative vitreoretinopathy (PVR) and cultured retinal pigent epithelium (RPE) cells. Methods Fifteen human epiretinal membranes were obtained from eyes undergone vitrectomy for rhegmatogenous retinal detachment complicated with PVR and observed by immunohistochemical examination to study the expression of HGFR. Using the immunohistochemical technique to evaluate the expression of HGFR in cultured RPE cells. Results In 6 membranes of PVR-grade C, HGFR were expressed in 5/6, and 7 cases were detected in 9 membranes of PVR-grade D.RPE cells express readily detectable levels of HGFR. Conclusion The findings indicate that HGF might be involved in the formation of epiretinal membranes in PVR. (Chin J Ocul Fundus Dis, 2002, 18: 221-223)

  • A comparative study of hepatocyte growth factor levels in the vitreous of patients with diabetic retinopathy

    Objective To determine the concentration of hepatocyte growth factor (HGF) in vitreous of diabetic retinopathy (DR) and investigate the result and influence of HGF in neovascularization in proliferative diabetic retinopathy (PDR). Methods The high sensitive sandwich enzyme-linked immunosorbent assay (ELISA) was used to measure the level of HGF in vitreous of normal group for 10 eyes, simple DR group for 7 eyes, PDR without iridal neovascularization group for 24 eyes, PDR with neovascularization of iris group in 9 eyes, other retinopathy group associated with neovascularization for 8 eyes. Results The mean value of HGF level in vitreous of the former groups was: (3.3 ±1.9) μg/L in normal group; (4.8±2.5) μg/L in simple DR group; (13.0 ±5.2) μg/L in PDR without iridal neovascularization group; (18.6±7.2) μg/L in PDR with neovascularization of iris group;(12.1±8.9) μg/L in other retinopathy associated with neovascularization group. Stastistics showed that HGF level in PDR group and other retinal diseases associated with neovascularization were significantly higher than that in normal group (P<0.01), PDR with neovascularization of iris group showed a higher mean vitreous HGF concentration than those in PDR without iridal neovascularization and simple DR group (P<0.05 or P<0.01). Conclusion Our results indicate that vitreous HGF may play an important role in retinal neovascularization in PDR and other retinal diseases. (Chin J Ocul Fundus Dis, 2002, 18: 131-133)

  • Effect of hepatocyte growth factor on migration and proliferation in cultured retinal pigment epithelial cells

    Objective To investigate the effect of HGF on proliferation and migration in cultured human RPE cells. Methods Human RPE cells cultured in serum-free medium were treated with HGF(1,2,10,50,100 mu;g/L), and MT T assay was used to detect the growth of the cells; an in vitro wound healing model was used to count the number of cells that had entered the denudate area in RPE mig ration treated with HGF (1,2,10,50,100 mu;g/L) after 20 h. Results HGF(10,50,100 mu;g/L) increased proliferation rates of cultur ed human RPE (18.2 % to 34.8 %), and at a concentration of 50 mu;g/L on day 3 HGF induced the maximal increase of proliferation (Plt;0.01); HGF showed effects on migration of 9.3 %, 113.0 %, 91.7 % and 50.3 % at the concentrations of 2,10,50, 100 mu;g/L, respectively. HGF stimulated t he best migratory response in RPE cells at a concentration of 10 mu;g/L (113 %). Conclusion HGF can induce the proliferation and obvious migration of RPE cells, consequently HGF was a mitogen and potent migratory factor for human cultured RPE cells.  (Chin J Ocul Fundus Dis, 2001,17:307-310)

  • P eroxisome proliferators-activated receptor gamma-agonists inhibis the expression of phosphorylation of signal transducer and activator of transcription 6 in T-lymphocytes of patients with acute asthma in vitro

    Objective To explore the possible anti-inflammatory mechanism of peroxisome proliferators-activated receptor(PPAR) gamma-agonists by investigating the effects of Rosiglitazone on the expression of phosphorylation of signal transducer and activator of transcription 6(p-STAT6) and the secretion of interleukin(IL)-4 in T-lymphocytes from patients with acute asthma.Methods Peripheral blood T-lymphocytes from 10 healthy volunteers(group A) and 10 patients with acute asthma were isolated,purificated and cultured.T-lymphocytes from the asthma patients were divided into a control group(group B) and a Rosiglitazone treated group(group C).Rosiglitazone was added with a single dose of 10-4 mol/L at 0 hour of cultrue.After cultured for 48 hours,the concentration of IL-4 in supernatant of each groups were detected by ELISA.The express of p-STAT6 in the T-lymphocytes were determined by Western blot and immunohistochemical techniques.Results The levels of IL-4 were increased markedly in group B than those in group A and group C[(170.34±9.05)pg/mL vs(76.82±7.06)pg/mL and(123.59±8.70)pg/mL,both Plt;0.01],and which in group C was significantly lower than group A(Plt;0.01).The levels of p-STAT6 in T lymphocytes were increased markedly in group B than in group A and C[Western blot:(6.28±0.19 vs 3.07±0.18 and 4.12±0.16;immunohistochemistry:(36.58%±7.41)% vs(11.39±4.02)% and(23.92±5.8)%,all Plt;0.01),and which in group C were significantly higher than that in group B(both Plt;0.01).There was a positive correlation between the level of p-STAT6 and IL-4(Plt;0.01).Conclusion The levels of p-STAT6 and IL-4 in T-lymphocytes of patients with acute asthma were suppressed by Rosiglitazone in vitro.

  • Meta-analysis of prevalence and risk factors of chronic obstructive pulmonary disease in China plateau

    Objective To analyze the prevalence rate of chronic obstructive pulmonary disease in people over 40 years old in China plateau (altitude≥1 000 m) in recent years. Methods PubMed, Embase, Cochrane Library, China National Knowledge Infrastructure, WanFang Data, VIP databases and Chinese Medical Database were searched, and the articles published between January 1, 2010 and December 5, 2022 were collected. Two reviewers independently reviewed the literature, extracted data, and assessed the risk of bias of included articles. Meta-analysis was performed with Stata 17.1. Results A total of 13 articles were included, with a total sample size of 37 230 people, including 3 697 chronic obstructive pulmonary disease patients. The prevalence rate of chronic obstructive pulmonary disease in people over 40 years old in China plateau was 9.0% [95% confidence interval (0.07, 0.11), P<0.001]. The results of subgroup analysis showed that males (12.4%), smokers (13.4%), 61-70 years old (15.1%), primary school and below (10.3%), low body weight (12.2%) and Yunnan (11.5%) had the highest prevalence rates within the subgroup (P<0.05). Conclusions The prevalence of chronic obstructive pulmonary disease in China plateau is still high. Gender, age, smoking, education level, body mass index and region are still important influencing factors of chronic obstructive pulmonary disease.
