  • Marrow Stroma Cell Culture and Identification of SD Rat in Vitro

    Objective To research the methods and techniques of SD rat bone marrow stromal cells (MSCs)culture in vitro and to provide a large number of MSCs for cell therapy. Methods Bone marrow from the femur and tibia of the early age SD rats was taken to culture to the passage 1-4 (P1-P4), its growth was observed and P3 cells were evaluated by HE staining and immunohistochemisty. Results The growing speed of P1-P3 was faster than that of P0, cells fusion was 85%-90% after 3-4 d and the cells were arranged in groups liked whirlpool shape or parallel; (3.4-3.6)×104/cm2 cells were gained and the total cell number of P1-P3 was 4.08×106, 2.44×107 and 2.85×108 respectively, the rate of trypan blue rejecting stained was 95%-97%. P4’s growing speed was slower than before, 1.42×109 in 3.0×104/cm2 cells were gained, and the rate of rejecting stained was 95%. P4-cell output was amplified nearly 2 000-fold higher than P0-cell. P3 immunohistochemical analysis indicated CD105+ cells 61.9%, CD44+ 45.4%, CD29+ 16.8%, CD45+ 8.2%, CD31+ 13.7%, CD34+ 8.3% and CD11b+ 1.5%, respectively. Conclusion The culture of whole bone marrow is suitable for a large number of MSCs provision in vitro, and can meet the needs of the cell therapy research.

  • Effectiveness of transverse tibial bone transport in treatment of diabetic foot ulcer

    ObjectiveTo evaluate the effectiveness of transverse tibial bone transport in treatment of diabetic foot ulcer. MethodsBetween June 2014 and December 2017, 17 patients with diabetic foot ulcer were treated. There were 11 males and 6 females, with a median age of 57 years (range, 46-72 years). The duration of diabetes was 2.4-32.0 years (mean, 16.0 years). According to the Wagner grading criteria, 7 cases were rated as grade 2, 8 cases as grade 3, and 2 cases as grade 4. The CT angiography (CTA) showed the arterial occlusion or stenosis of varying degrees below knee joint. All cases were treated with transverse tibial bone transport. Bone transport started at 3 to 5 days after placing external fixator and lasted 14 days (1 mm per day), and then reverse transport started. The total transport time was 28-30 days. ResultsAll 17 patients were followed up 5-12 months (mean, 8.5 months). During transportation, 4 cases had screw orifice infection, and 3 cases had liquefaction and seepage. And other 15 cases ulcers healed and the healing time was 35-72 days (mean, 48 days). There were significant differences in visual analogue scale (VAS) scores, skin temperature, ankle brachial index (ABI) between before and after ulcer healed (P<0.05). The CTA showed that the collateral circulation was established. The foot ulcer of 2 cases (Wagener grade 3 in 1 case and grade 4 in 1 case) still progressed after treatment, and amputation was performed. ConclusionTransverse tibial bone transports has good effectiveness for diabetic foot ulcer.

  • The detection rate of maternal postpartum depression in China: a meta-analysis

    ObjectiveTo systematically evaluate the detection rate of postpartum depression in Chinese maternal population and to provide a scientific basis for the prevention and treatment of postpartum depression.MethodsWe searched CNKI, WanFang Data, VIP, PubMed, EMbase and The Cochrane Library databases to collect studies on the detection rate of postpartum depression in Chinese maternal population from January, 2001 to August, 2019. Two reviewers independently screened literature, extracted data and evaluated the risk of bias of included studies. Meta-analysis was performed using Stata15.0 software.ResultsA total of 24 studies involving 38 357 cases were included. Meta-analysis results showed that the total detection of postpartum depression in Chinese females was 15% (95% CI 12% to 17%). Subgroup analysis showed that the detection of postpartum depression was 12.3% (95% CI 9.3% to 15.2%) in the south and 17.3% (95%CI 12.1% to 22.5%) in the north. According to the Edinburgh postpartum depression scale, the rate was 14.5% (95%CI 11.4% to 17.5%); and for other scales, the rate was 15.0% (95% CI 8.9% to 21.2%); simple random sampling method was 14.8% (95%CI 11.0% to 18.7%), and cluster sampling was 16.3% (95%CI 12.0% to 20.5%). The rate was 15.8% (95%CI 9.3% to 22.3%) from 2001 to 2010, 13.5% (95%CI 7.7% to 19.2%) from 2011 to 2014, and 14.8% (95%CI 10.9% to 18.6%) from 2015 to 2019. Sensitivity analysis showed that the combined results were stable.ConclusionsThe detection rate of postpartum depression in Chinese maternal population is high, and early screening and related intervention should be paid more attention to these population.

  • Efficacy of Reteplase and Ateplase in Treatment of Acute Massive Pulmonary Thromboembolism: A Comparative Study

    Objective To compare the efficacy of reteplase and ateplase in the treatment of acute massive pulmonary thromboembolism ( PTE) in emergency. Methods From January 2005 to December 2009,42 patients with acute massive PTE were treated by intravenous thrombolysis with reteplase or ateplase. The thrombolysis efficacy, bleeding incidence and mortality were measured. Results In the reteplase group, the emergency thrombolysis effective rate was 88. 9% among 18 patients. Mild bleeding occurred in 3 patients,moderate bleeding in 1 patient, and 2 cases died in hospital. In the ateplase group, the emergency thrombolysis effective rate was 75% among 24 patients. Mild bleeding occurred in 3 patients, moderate bleeding in 2 patients, and 3 cases died in hospital. The thrombolysis effective rate, bleeding incidence and mortality had no significant difference between the two groups. Conclusion Both the reteplase and ateplase thrombolysis therapy are safe and effective in the treatment of acute massive PTE, but reteplase thrombolysis therapy is more convenient in emergency.

  • Cardiovascular collapse caused by balloon pre-dilation during transcatheter aortic valve replacement: a case report

    Circulatory collapse is a common complication of transcatheter aortic valve replacement (TAVR), mainly due to new severe aortic regurgitation after balloon pre-dilation. This article reports the case of an 80-year-old female patient with severe aortic stenosis, who had a type 1 bicuspid aortic valve, with calcified aortic valve commissure between the right and left coronary cusps. During the procedure of TAVR, severe aortic regurgitation was caused by pre-dilation with 20 mm and 23 mm balloons. Then circulatory collapse and cardiogenic shock occurred. After the emergency deployment of the Venus A L26 valve, the cardiovascular hemodynamics was immediately improved. This case suggests that oversized balloon dilation should be avoided to prevent circulatory collapse caused by massive aortic regurgitation.

  • Study on the reporting checklist of technical specifications for traditional Chinese medicine

    Technology in traditional Chinese medicine (TCM), which has a long history, provides excellent traditional culture and valuable spiritual wealth in China. TCM standardization technology will provide an important basis for guiding and evaluating the professional level and service quality of technicians in TCM. And it is also conducive to the sustainable development of TCM technology. However, at present, there is no relevant research on the development process of technical specifications for TCM. So, the urgent need is how to develop standardized technical specifications, and apply them to patients in practice. Therefore, this study first convened relevant experts, including TCM clinical experts, epidemiologists, methodology experts, etc., to form the core expert group, implementation group and quality assurance task group. According to the personnel functions, they are divided into technical specification steering committee, consensus expert group, secretariat group, external review group, evidence evaluation group, etc. Then, the initial entries are collected by systematically searching the current TCM technical guidelines, consensus and specifications and referring to the existing technical specifications. Finally, expert opinions were collected based on the Delphi survey, and the final reporting checklist of technical specifications for TCM was formed after consensus discussion. The study can provide evidence-based methodological guidance for the development of TCM technical specifications, and promote the standardization and internationalization of TCM technical specifications.
