  • The Effects of Newcastle Disease Virus on Tumor Infiltrating Lymphocyte

    ObjectiveTo study the effects of newcastle disease virus (NDV) on tumor infiltrating lymphocyte (TIL). MethodsTILs were dissociated from 6 cases of primary hepatic carcinoma. Each case was divided into 3 portions equally, and was added 20 μl NDV with HA titer of 1∶160 (NDV group), rIL2 with the end concentration of 1 000 u/ml (rIL2 group) and 0.9% sodium chloride (control group) respectively, then cultured in the condition of 37℃,5% CO2. CD3, CD4 and CD8 were assayed by flow cytometry. The contents of IL2, IFNγ and TNFβ in supernate of NDV group and control group were examined by ELISA.ResultsCoupled material revealed that CD3 and CD4 were significantly higher in NDV group than in the other two groups (P<0.05, P<0.01). CD8 had no significant changes in three groups. The contents of IL2, IFNγ and TNFβ in NDV group were significantly higher than those in control group (P<0.01, P<0.01 and P<0.05 respectively). ConclusionNDV can activate TIL directly.

  • STUDY ON ANTITUMOR ROLE OF NEWCASTLE DISEASE VIRUS COMBINED THERMIC SOLIDIFIED TUMOR VACCINE IN RATS

    In this study, the role of newcastle disease virus (NDV) combined thermic solidified tumor vaccine in inhibiting growth of tumor and immune control was investigated, and rate of inhibiting tumor and cellular immunity were measured. The results showed that rate of inhibiting tumor in experimental group Ⅰ and Ⅱ were 24.8% and 41.1% respectively; average weight of tumor was significantly lower in both experimental groups than in control group, and activity of natural killing (NK) cells in experimental groups was higher than that in control group (P<0.01). This suggests that NDV combined thermic solidified tumor vaccine can inhibit growth of tumor and improve activity of NK cells, and their effects are better than that of NDV.

  • Effects of Newcastle Disease Virus and Adriamycin on Surface Structure and Actin of Hepatocellular Carcinoma Cell Lines SMMC-7721

    【Abstract】ObjectiveTo compare the effects of newcastle disease virus (NDV) and adriamycin (ADM) on surface structure and actin of hepatocellular carcinoma cell lines SMMC-7721. Methods SMMC-7721 carcinoma cell lines were divided into 2 groups. NDV was added into one group, while ADM was added into the other group. The cells were then cultured at 5 time phases (8, 16, 24, 36 and 48 h). Intracellular actin and Ca2+ were examined by using immunofluorescence method. CD44 and intercellular adhesion molecule-1 (ICAM-1) were detected by using immunochemical method and flow cytometry, respectively. The change of cellular surface structure was observed by scan electron microscope. Results Cells gradually contracted and turned round over time. It was observed that actin was segmented and cells alignment became disordered. The mean fluorescence intensity of actin decreased in both groups, but it was obvious in NDV group. There were significant differences of fluorescence intensity between 2 groups at the phases of 16 h (P<0.05), 24 h (P<0.05), 36 h (P<0.01) and 48 h (P<0.05), except the one after 8 h. Intracellular Ca2+ concentration increased gradually in both groups, and the amplifications in NDV group were significantly higher at the phases of 24 h, 36 h and 48 h than those in ADM group (P<0.01, P<0.05 and P<0.01, respectively). There were also differences at 8 and 16 h, but there were no statistical significance. The expression of CD44 in cells decreased. The mean fluorescence intensity of ICAM-1 raised gradually, and then came to peaking at 36 h, but there was no significant difference between two groups. All the above indices between different phases in the same group showed significant differences (P<0.05). Conclusion Both NDV and ADM could make tumor cells degenerate and rupture, but the effect of NDV is more intensive. It could increase the fragility of cells and hasten the process of cell rupture. Disintegrated cancer cell and changes of adhesion molecule could lead cancer cells be identified, encapsulated, and killed by immune cells under static condition.

  • Study on Anti-Gastric Cancer Effects Induced by NDV-ATV and Dendritic Cells

    【Abstract】Objective To explore the effect against gastric cancer induced by Newcastle disease virus modified autologous tumor vaccine (NDV-ATV)pulsed dendritic cells(DCs). Methods The Newcastle disease virus infected the gastric cancer lines (MNK45) and was lost its activity. Peripheral blood mononuclear cell (PBMC) were cultured under condition of recombinant human granulocyte macrophage-colony stimulating factor (1 000 u/ml)+IL-4(1 000 u/ml) + TNF-α(100 ng/ml). The tumor antigen specific cytotoxic T lymphocytes (CTL) was generated from activated autologous T cell by the Newcastle disease virus infected the MNK45 pulsed DC. And Cyto Tox 96TM in vitro assayed the cytotoxicity of CTL to MNK45. Thawed gastric cancer cell antigen were used as control in these experiments. Results The killing rate of MNK45 by antigen specific CTL reached (90.15±9.82)%, which was nearly twice as high as that of control(60.57±5.74)%. The CTL had much higher cytotoxicity to different differentiated type of gastric cancer cells such as MGC803〔(52.23±6.45)% 〕 and SGC7901〔 (61.75±8.84)%〕, as compared with LOVO〔(9.11±3.42)%〕 and HepG2 〔 (8.30±3.12)%〕tumor cells(P<0.05). Conclusion Efficient and specific of against gastric cancer immunoreaction can be induced in virtue of NDV-ATV pulsed DCs, NDV-ATV loaded DCs might provide a new kind of theraputic means for gastric cancer.
