  • EXPERIMENTAL STUDY ON THE TREATMENT OF RAT ACUTE HEMORRHAGIC NECROTIZING PANCREATITIS WITH MANNITAL

    This study aimed to assess the therapeutic effect of mannital administration on acute hemorrhagic necrotizing pancreatitis (AHNP), 28 Wistar rats were randomily divided into therapeutic group and control group after induction of AHNP by retrograde intraductal injection of 5 percent sodium taurocholate. The rats of therapeutic group received intravenous 20% mannital (1g/kg) through tail vein, once in 12 hours, until the end of experiment; control group received saline (5.0 ml/kg) with the same way. Blood of all the rats were collected from heart and the rats were killed after 96 hours. Results: lipid peroxide (LPO) in pancreatic tissue, LPO in serum, lactate dehydrogenase (LDH), alpha-1-antitrypsin (α1-AT), glutamicoxalacetic transaminase (GOT), necrotizing square of pancreatic tissue in the therapeutic group were significantly less than those in control group (P<0.05 or P<0.01). The damage to pancrease, heart, liver, kidney in the therapeutic group were lighter than those of the control group and the mortality was lower (P<0.05).Conclusions: Mannital can scavenge the oxygenderived free radicals and play a therapeutic role in AHNP.

  • STUDY ON GASTRIC MUCOSAL INJURY INDUCED BY SHOCKREPERFUSION IN PORTAL HYPERTENSION RATS WITH CIRRHOSIS

    Electron spin resonance (ESR) spectroscopy and spin trapping agent PBN were applied to measure directly the changes of oxygen free redicals (OFR) in gastric mucosa of rats with portal hypertension (PHT) injured by shockreperfusion, and treated with superoxide dismutase (SOD), radix salviae miltiorrhizae (RSM), with concomitant monitoring activity of SOD and pathology of gastric mucosa. Results showed that the amount of OFR increased markedly in gastric mucosa of PHT rats during the shock-reperfusion. The pathological changes were in accordance with alteration of the amount of OFR and the activity of SOD. Gastric mucosa in PHT was more susceptible to shock-reperfusion insult than normal controls. The anti-oxidant SOD, RSM used at early stage exerted mild gastric mucosal insult through different mechanisms.

  • OXYGEN FREE RADICAL AND HUMAN FIBROUS CAPSULE

    The specimens were obtained from 7 fibrous capsules two years after mammoplasty with silicone prosthesis, and were examined by electron spin resonance (ESR) andmicroscope. The result showed that a marked quantities of oxygen free radical existed in the capsules and that the main pathological change was the infiltration of massive inflammatory cells and deposition of collagen. It was suggested that the fibrous capsule formation was concerned with the oxygen free radical. The origin of the oxygen free radical in mammoplasty with slicone was also discussed.

  • DETECTION OF FREE RADICALS IN FIBROUS CAPSULES

    Determination of free radical has been done in the fibrous capsules with electron spin resonaee (ESR). The results showed that there was a marked production of oxygen free radical, and the superoxide dismutase could scavenge it effectively. The author discussed the oxygen relationship of free radicals with the collagen synthesis in the fibrous capsule.

  • EXPERIMENTAL STUDY OF PREVENTIVE EFFECT ON FORMATION OF CATARACT IN DIABETIC RATS

    We assayed the levels of free radical and scavenger in the blood and lens of streptozotocin-in-duced diabetic SD rats, and found that the levels of lipoperoxide(LPO),MDA were higher than that of normal SD rats, and the total superoxygen dismutase (T-SOD), Cu-Zn-SOD) were lower that that of normal rats ( P lt;0.01 ). Simultaneous injection of streptozotocin and large dose of SOD could no avoid the occurence of diabetes mllitus, but did improve the metabolism of free radical in blood and lens. Hence, we think that large dose of SOD might be effective in preventing to development of diabetic cataract which is related to deterioration of free radical metabolism.   • Protective Effect of Resveratrol on Intestinal Mucosal Ischemia-Reperfusion Injury in Rats with Severe Acute Pancreatitis

    Objective To observe the influence of resveratrol on superoxide dismutase (SOD), malondialdehyde (MDA), intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) of intestinal mucosal ischemia-reperfusion injury protection in rats with severe acute pancreatitis (SAP). Methods Fifty-four rats were divided into three groups randomly: sham operation group (SO group), SAP model group (SAP group) and resveratrol-treated group (Res group). SAP model was made by injecting sodium taurocholate 50 mg/kg to pancreatic bile duct and resveratrol was given intravenously at 5 min after inducing SAP model. The rats were sacrificed at 3 h, 6 h and 12 h after inducing SAP model respectively by equal number. The levels of MDA, SOD, ICAM-1 and VCAM-1 and histological changes of small intestine were measured. Results The level of MDA in small intestine tissue in SAP group was significantly higher than that in SO group (P<0.05), while the activity of SOD was significantly lower in the relevant tissues (P<0.05). The expressions of ICAM-1 and VCAM-1 in SAP group were higher than those of SO group (P<0.05). The activity of SOD in small intestine tissue in Res group was significantly higher than that in SAP group (P<0.05); while the level of MDA was significantly lower in the relevant tissues (P<0.05). The expressions of ICAM-1 and VCAM-1 in Res group were lower than those of SAP group (P<0.05). Conclusions Oxygen free radicals are concerned with the process of pathological changes in intestinal mucosal ischemia-reperfusion in rats with SAP. Resveratrol might increase SOD activity and decrease MDA level to attenuate lipid peroxidation in small intestine of SAP, and reduce the expressions of ICAM-1 and VCAM-1 in intestine, thus diminish the damage of the intestine in SAP. And it acts as a protective effect to small intestinal ischemia-reperfusion injury.

  • Antioxidant Function of Solanesol and Its Inhibitory Effect on Tyrosinase

    The present paper intends to discuss the antioxidant and tyrosinase inhibition effect of solanesol from three aspects of ultraviolet radiation and free radical scavenging. The paper makes a survey on diurnal variation rule of the minimum ultraviolet transmittance and ultraviolet transmittance of solanesol, hydroxyl (·OH) free radical scavenging method of Smirnoff reaction system model, superoxide anion O2-· free radical scavenging method of pyrogallol autoxidation, and the inhibitory effect of solanesol on tyrosinase activity by enzyme kinetic method. The experiment results showed that solanesol could effectively scavenge lipid radicals to block lipid peroxidation, and inhibit effects on tyrosinase. Solanesol is a natural extract which could be used to prevent senile atrophy of human skin and senile plaque.
