  • Expression of p27 in Hepatocellular Carcinoma and Its Clinical Significance

    ObjectiveTo study the expression of p27 in hepatocellular carcinoma (HCC) and its clinical significance.MethodsFortyfive specimens of HCC and 30 specimens of adjacent noncancerous lesions obtained from 45 patients who underwent surgery were examined for p27 expression by immunohistochemistry SABC method. The diameter of tumor ranged from 1 cm to 19 cm (d ≤5 cm, 9 samples; 5 cmlt;d≤10 cm, 19 samples; d>10 cm, 17 samples). These tumors were graded according to the criteria described by EdmondsonSteiner: highdifferentiated HCC group (Grade Ⅰ+Ⅱ), 26 samples; lowdifferentiated HCC group (Grade Ⅲ+Ⅳ), 19 samples. According to the clinicopathological features: 19 samples were poorly encapsulated, 15 samples had portal invasion, 11 samples had extrahepatic metastasis, 12 samples had intrahepatic metastasis; all of the above were classified as the invasive and metastatic group, while the others were classified as the noninvasive and nonmetastatic group. ResultsThe average labeling index (LI) of p27 in HCC was significantly higher than that of adjacent noncancerous lesions (45.87±14.21 vs 33.77±12.92, Plt;0.001). The LI of p27 in lowdifferentiated HCC group (stage Ⅲ+Ⅳ) was significantly lower than that of highdifferentiated group (stage Ⅰ+Ⅱ), 34.46±12.29 vs 52.80±11.36 (Plt;0.001). The LI of p27 had significant difference between the large ones (d>10 cm, 37.59±13.12) and the small ones (d≤5 cm, 53.28±15.17 or 5 cmlt;d≤10 cm, 49.50±10.96) (Plt;0.05). The LI of p27 in the invasive and metastatic group was significantly lower than that in the noninvasive and nonmetastatic group (41.42±12.86 vs 51.44±14.10, Plt;0.05).ConclusionThe expression of p27 is more frequently detected in HCC than in adjacent noncancerous lesions. It indicates that p27 might be a compensatory factor during HCC carcinogenesis. The LI of p27 significantly decreases in poor differentiation group, invasive and metastatic group. It indicates that p27 might be related with the differentiation, invasion and metastasis of HCC.

  • EXPRESSION AND SIGNIFICANCE OF CYCLIN D1, CYCLINE AND P27 IN GASTRIC CARCINOMAS

    Objective To study the expression and its significance of cyclin D1,cyclin E and p27 protein in gastric carcinoma. Methods The expression of cyclin D1,cyclin E and p27 protein in 60 cases of gastric carcinoma were detected by immunostaining method. Results The positive rates of cyclin D1,cyclin E and p27 protein expression in gastric carcinoma were 41.7%(25/60),38.3%(23/60)and 36.7%(22/60),respectively.The expression of cyclin D1 was significantly correlated with Lauren type of tumor,the expression of cyclin E was significantly correlated with tumor differentiation,infiltration and stage,while the expression of p27 protein was significantly correlated with tumor infiltration,lymph nodes metastasis and stage. Further more, the expression of cyclin E was inversely correlated with the expression of p27. Conclusion Detection of cyclin D1,cyclin E and p27 expression in gastric carcinoma may be helpful in determining tumor progression.

  • THE SIGNIFICANCE OF EXPRESSION OF P27 IN THYROID FOLLICULAR TUMORS

    【Abstract】Objective To study the significance of expression of p27 in thyroid follicular tumors. Methods The tumor samples were obtained from 53 thyroid follicular tumors, and 10 normal thyroid tissues were used as a control group. p27 was detected by the immunohistochemical staining S-P method. Results The normal thyroid group had the highest expression of p27.There were significant differences in the expression of p27 between thyroid follicular adenomas (FA) and follicular carcinomas (FTC), FA had much higher expression of p27 than FTC did. There were no significant differences in the p27 staining indexes between minimally and widely invasive FTC nor between metastized and non-metastized FTC. Logistic regression showed that p27 were effective in distinguishing FA from FTC(P=0.0048). Conclusion It appears that p27 may be necessary for the malignant transformation of the thyroid follicular epithelium, which might relate to a decrease in p27. p27 detection may be helpful in distinguishing FA from FTC.

  • Expressions of S-Phase Kinase-Associated Protein 2 and p27 Protein in Gastric Carcinoma and Pericancerous Tissues

    Objective To study the expressions of SKP2 and p27 in gastric carcinoma and pericancerous tissues and to detect the relationship between their expressions and clinicopathological features. Methods Forty-nine cases of gastric carcinoma spicemen and 20 cases of tissue adjacent to the carcinoma were cut and made into paraffin-embedded slices. The expressions of SKP2 and p27 were then detected by SP immunohistochemical method. Results The positive expression rate and score of SKP2 were both significantly higher in the gastric carcinoma tissues than those in pericancerous tissues (P<0.01), whereas those terms of p27 were higher in pericancerous tissues (P<0.05, P<0.01). It was observed that the pericancerous tissues with positive SKP2 expression or with negative p27 expression showed atypical hyperplasis ranging from moderate to severe degrees. The positive rate and score of SKP2 were significantly lower in the cases of infiltrating depth T1+T2, without-metastasis of lymph node, with-metastasis of the first site lymph node, and without-metastasis of distant organs than those in infiltrating depth T3+T4, with-metastasis of lymph node, with-metastasis of the second or third site lymph node and with-metastasis of distant organs in gastric carcinoma tissues (P<0.05, P<0.01), whereas the results were contrary for p27 (P<0.01). There was a negative correlation between the score of SKP2 and that of p27 in gastric carcinoma tissues (r=-0.65, P<0.01). Conclusion The expressions of SKP2 and p27 may act as important biological markers to reflect carcinogenesis, progression, biological beheviors and prognosis of gastric carcinoma.
